Background | DSPE-PEG-SH, MW 1,000 is a thiol functionalized PEG lipid which is reactive with maleimide to form a stable thioether bond. The phospholipid PEG can be used to prepare liposome or lipid nanoparticles for delivering nutrients/therepeutics/drugs, such as liposomal anti cancer drug/vaccine (mRNA or DNA vaccine). 1,2-distearoyl-sn-glycerin-3-ethanolamine phosphate (DSPE) conjugated polyethylene glycol, DSPE peg is a kind of phospholipid peg conjugate with hydrophilic and hydrophobic properties. Pegylated phospholipids are excellent liposome forming materials and can also be used for drug delivery, gene transfection and vaccine delivery. PEGylation of phospholipids significantly improved the blood circulation time and stability of encapsulated drugs. These materials can also be used for targeted drug delivery by modifying their surfaces with targeted ligands (e.g., antibodies, peptides). Dspe-peg-sh is an active phospholipid peg reagent with thiol group at the end of another peg. DSPE (1,2-di-stearoyl-sn-glycerin-3-phosphate ethanolamine) is a saturated 18C phospholipid, which is often used in the synthesis of liposomes. Polyethylene glycol (PEG) coupled DSPE is hydrophilic and can be used for drug delivery, gene transfection and biomolecular modification. PEGylation of phospholipids can significantly improve the blood circulation time and stability of capsule drugs. Sulfhydryl group (dspe-peg2000-sh) and maleimide (dspe-peg2000-mal) can easily form stable thioether bond in ph6.5-7.5. Mercaptan group has high affinity for gold surface, which is widely used in gold nanoparticles or gold film modification. PEG modification can improve solubility and stability, and reduce immunogenicity of proteins and peptides. It can also inhibit the nonspecific binding of charged molecules on the modified surface. |
